South Australia is facing a raft of new coronavirus bans and border restrictions, as authorities launch a rapid response to extinguish a feared second COVID-19 wave.

"It is fair to say that we are facing our biggest test to date," Premier Steven Marshall warned.

As the other states closed to SA and more troops were called in, several schools closed and health alerts were issued for swathes of the northern suburbs.

The number of people linked to the Parafield family cluster rose to 16 confirmed cases, plus a further three children who are being treated as active cases but are yet to be confirmed.

Overall the state recorded 18 new COVID-19 cases on Monday, for a total of 34 active cases

The cluster's source is a female cleaner, aged in her 50s, who worked at the Peppers medi-hotel in Waymouth Street, before she unwittingly triggering the state's first community transmission case in more than six months.

The cleaner, believed to be from a large Indian family, is thought to have infected her husband, aged in his 60s, and her two elderly parents.

The pair, both aged in their 80s, were on Monday listed in a stable condition in a special Royal Adelaide Hospital COVID ward as a precaution due to their ages.

As of Monday night, at least 167 close contacts were being checked for symptoms. Dozens more are in Adelaide medi-hotels, while a further 96 police officers were isolating. A worker at Yatala prison has also tested positive, sparking concerns about the state's jail system.

South Australia's virus outbreak has forced thousands of people into quarantine as the state deals with a cluster that has more than doubled.

South Australia's virus outbreak has forced thousands of people into quarantine as the state deals with a cluster that has more than doubled.

As a precaution, there are now around 4000 people considered close contacts who have been quarantined, chief health officer Professor Nicola Spurrier revealed this afternoon.

"These people have been advised to quarantine and they are being contacted on a daily basis for a symptom check," Prof Spurrier said.

"This is a huge amount of work, as you can imagine, to make sure that we are in regular contact with these people."

There are now 21 cases linked to the Parafield {INDIAN} family cluster with 15 of those coming from the same family.

The family cluster includes three young children, one of which is a one-year-old-baby, Premier Steven Marshall confirmed this morning.

Overall the state recorded 18 new COVID-19 cases yesterday and five new cases today, for a total of 34 active cases, including hotel quarantine cases.

At least three of the 34 active cases are employees at the Anglicare Brompton Aged Care Home in Adelaide, which the CEO of Anglicare SA this morning described as his "worst nightmare".

All residents have so far tested negative however CEO Peter Sandeman said they would be tested again tomorrow and any close contacts were being isolated.

SA Health is continuing to study the genomic strains of each of the state's new cases, which will help them determine who caught the infection first.

A female {INDIAN} cleaner in her 50s, working at one of Adelaide's medi-hotels named Peppers, is believed to have caught it first, later inadvertantly passing it onto their family and two unrelated hotel security guards.

The cluster has triggered closures and isolation warnings across the state with five schools shutting down for deep cleaning and hundreds of people being ordered into isolation.

Testing clinics are also buckling under the thousands of people wanting to get tested, with wait times at some of Adelaide's sites blowing out to 11 hours.
